Reducing costs of whole genome sequencing from the extraction stage

Ancient DNA is often only preserved in minute amounts in archeological material such as fossilized bones and teeth. Making sure that all ancient DNA molecules preserved are effectively extracted is, thus, essential in any paleogenetic study, all the more than extraction procedures are generally destructive.

A new article published by CGG researchers in the journal Molecular Ecology Resources benchmark the three most commonly used extraction methods in ancient DNA research. Cristina Gamba and colleagues show that column-based methods outperform in-solution methods, providing higher-quality DNA and reducing the costs underlying whole genome sequencing of ancient individuals and/or extinct species. The two column-based methods tested allow the retrieval of even the most degraded category of ancient DNA molecules and show similar efficacy and hands-on time. One, however, appears 40% cheaper and should, thus, be recommended.
